Cucumber Green Mottle Mosaic Virus Diagnostic Work Instructions
APHIS’ Plant Pathogen Confirmatory Diagnostics Laboratory (PPCDL), formerly called the Beltsville Laboratory, completed two Cucumber
green mottle mosaic virus (CGMMV) diagnostic work instructions for screening laboratories:
- WI-B-T-2-19 Homogenization of Cucurbit Seeds for Enzyme-Linked Immunosorbent Assay (ELISA) and for Ribonucleic Acid (RNA) Extraction for Cucumber Green Mottle Mosaic Virus Detection
- WI-B-T-2-19 describes the subsampling and grinding procedure of cucurbit seed samples for ELISA and RNA extraction for CGMMV detection. This method was tested on cucumber, watermelon, cantaloupe/melon, squash, and pumpkin seeds.
- WI-B-T-4-4 ELISA for Screening Detection of Cucumber Green Mottle Mosaic Virus
- WI-B-T-4-4 describes a serological method for detecting CGMMV in both seeds and plants using an ELISA method from Agdia Inc. The method has been successfully tested with more than 40 CGMMV isolates from different geographical areas and is comparable to the International Seed Testing Association’s approved ELISA method.
